Antengene Updates Clinical Pipeline at Evercore Biotech Summit

Antengene Corporation unveiled fresh clinical data for its CLDN18.2 antibody-drug conjugate, ATG-022, during the Evercore 2nd China Biotech Summit. The company reported sustained therapeutic safety and long-term survival signals in gastric cancer patients, while detailing the expansion of its proprietary T-cell engager engineering platforms.

The latest results from the Phase II CLINCH study highlight the potential of ATG-022 in treating gastric cancer. Among patients receiving the 1.8 mg/kg dose, the study recorded an objective response rate of 46.7% with a median overall survival that has not yet been reached after a median follow-up of 14.03 months. Clinical safety data remains stable; despite six months of ongoing treatment, the incidence of Grade 3 or higher adverse events in the 1.8 mg/kg cohort rose only slightly to 21.0%. This profile supports the company's strategy to advance ATG-022 into first-line combinations with chemotherapy and anti-PD-1 antibodies.

Beyond current clinical assets, Antengene is scaling its T-cell engager (TCE) technology. The firm showcased its new TriGager platform, a tri-specific system capable of executing logic-gate functions such as AND-Gate and OR-Gate configurations. These mechanisms allow for more precise target recognition, potentially reducing off-target toxicity. This system complements the existing AnTenGager platform, which has already facilitated exclusive licensing agreements for candidates like ATG-201 and ATG-106 with UCB and K2 Therapeutics, respectively. Additionally, the company introduced ATG-207, a first-in-class bifunctional fusion protein currently under investigation for autoimmune disorders.
